Calibration and Reference Dyes


CAL Fluor® and Quasar® dye calibration standards are designed to improve the accuracy of sample signal in real-time thermal cyclers that require spectral calibration. They enable the instrument to store the fluorescent profile of each dye and control for channel cross-talk. Cross-talk is contamination of sample signal due to bleed-through from an adjacent spectral filter, an issue of particular importance in a multiplexed assay.

To achieve this, the dyes are formulated to resemble the experimental conditions. The native dye is covalently linked to the 5’end of a thymidine chain, 10 nucleotides long (T10) to better mimic the dye attached to a fluorescent probe. Using the dye calibration as a reference, the analysis software anticipates how much fluorescence to expect from each dye during amplification, and will subtract out signal from inappropriate channels. Calibration with the T10-dye standards decreases the likelihood of channel cross-talk.
