Plexor primer technology for real-time, quantitative PCR is a new and novel technique that requires only two primers for sensitive and specific quantitation of amplified DNA. The Plexor Technology is available from Promega Corporation of Madison, Wisconsin, Plexor primer technology takes advantage of the highly specific interaction between two modified nucleotides, isoguanine (iso-dG) and 5'-methylisocytosine (iso-dC) which form a unique base pair when incorporated in double-stranded DNA and pair only with each other.

In Plexor reactions, one PCR primer is synthesized with an iso-dC residue and a fluorescent label at the 5' end. The second PCR primer is unlabeled. Iso-dGTP nucleotides, modified to include a dabcyl quencher, are included in the PCR reaction mix. During the amplification reaction only dabcyl-iso-dGTP can be incorporated at the position complementary to the iso-dC residue. Incorporation of the dabcyl-iso-dGTP in close proximity to the fluorescent label effectively quenches the fluorescent signal.

The Plexor primer design software selects the appropriate fluorescent labels for single and multiplex PCR or quantitative reverse-transcription PCR (qRT-PCR) based on the optics of many popular real-time instruments.

The Plexor primer design software will send you an email with the target name, amplicon size and amplicon location used to generate each of the Plexor primers. Following this information the unlabeled primer and the iso-dC-containing primer along with fluorophore compatible with your specific real-time instrument.

Next, any off-the-shelf Plexor Human Control Gene Primer Sets which were selected will be listed.

Because your real-time instrument may need to be calibrated for the appropriate fluorescent dye linked to your Plexor primer, Biosearch Technologies will include the appropriate Calibration Dye upon request at no charge with your first Plexor primer order. If you require additional calibration dyes, these can be ordered directly on the Biosearch website.
