The I-Dig modification incorporates a digoxigenin internally or at the 5' terminus of an oligonucleotide. Digoxigenin is a steroid isolated from the digitalis plant used as a label in a variety of fluorescent, chemiluminescent, and colorimetric assays. As these plants are the only natural source of digoxigenin, the anti-DIG antibody binds with high specificity within a biological matrix. This hapten is conjugated to a modified thymidine which may participate in hybridization.
