Developing mRNA-based therapeutics requires rapid, high-quality RNA synthesis. In vitro transcription (IVT) is a method for synthesising RNA that has been instrumental to many of the advancements in RNA-based therapeutics.
The enzymatic generation of mRNA has several steps: plasmid cloning, plasmid linearisation, in vitro transcription, 5′ capping and adding a poly(A) tail. Purity and integrity of synthesised mRNA are critical to ensuring translational efficiency and reducing immunogenicity risks.

Analytical tools for characterising mRNA and other RNA-based therapeutics
- Hybridase Thermostable RNase H enables site-specific cleavage of RNA and is a great enzymatic tool for cleaving mRNA and long sgRNA prior to LC/MS analysis.
- RNA 5' Polyphosphatase generates 5'-monophosphate ends on uncapped synthesized RNA and works with Terminator 5'-Phosphate-Dependent Exonuclease for mRNA capping efficiency assays.
- Terminator 5'-Phosphate-Dependent Exonuclease degrades uncapped, 5' monophosphated RNA generated by RNA 5' Polyphosphatase.
- RNase A degrades ssRNA at C and U residues and can be used with 5' end DNA probes in mRNA digestion reactions to characterize capping by gel electrophoresis.
- T4 Polynucleotide Kinase, Cloned dephosphorylates certain oligoribonucleotides and is often added to digestion reactions to produce uniform termini, simplifying LC/MS analysis.
