3-ThioC6 is used to functionalize the 3' terminus of an oligonucleotide with a thiol group. Thiol modifications have become significant in the production of diagnostic probes. The free thiol can be used as a reactive handle to attach fluorescent dyes, maleimide compounds or for conjugation to proteins through disulfide linkages. The oligonucleotide will be shipped as a 3' disulfide with the form HO-C6-S-S-C6-oligo. To reduce it to a free thiol use 100 mM DTT and 0.2 M phosphate buffer pH 8.5 at room temperature for 30 minutes. The product can then be purified with a G-25 column.
