Sample collection

Samples are collected via at-home testing kit or by trained healthcare professional and submitted to a lab for processing. The Biosearch Technologies' workflow supports processing multiple sample types:

  • Nasopharyngeal swab
  • Oropharyngeal swab
  • Combined throat and nose swabs (CNTS)
  • Saliva

Viral RNA purification

Nucleic acid is found in all cells and viruses. DNA and RNA are types of nucleic acid. PCR testing looks for the presence of nucleic acid sequences specific to SARS-CoV-2.

To start the process, the nucleic acid must first be extracted or purified from the sample. Biosearch Technologies' oKtopure™ automated high-throughput extraction system and SARS-CoV-2 qualified sbeadex™ viral RNA purification kit speed this stage up.

Assay

In order to start the chemical reaction used to determine whether the target virus is present, a variety of reagents are added to the extracted nucleic acid.

These include Biosearch Technologies' RapidDxFire™ 1-Step RT-qPCR System, 2019-nCoV CDC-qualified Probe and Primer Kits for SARS-CoV-2, and Seracare AccuPlex™ SARS-CoV-2 Verification Panel, all verified for SARS-CoV-2 detection.

Amplification and detection

Now the chemical reaction can begin! The assembled reaction undergoes thermal cycling, causing the nucleic acid in the sample to be amplified via the polymerase chain reaction (PCR). This creates millions of copies of the viral nucleic acid, allowing it to be analysed.

The probes and primers that were added bind with the virus nucleic acid during the PCR reaction. If the virus is present, the probe will emit a fluorescent signal. Biosearch Technologies' automated IntelliQube™ System with Array Tape™ performs the PCR reaction, and then detects and reads the fluorescent signal, indicating a positive result. The IntelliQube platform is easily configurable and offers dual operation modes: qPCR (up to 4,500 tests per day) and end-point PCR (up to 35,000 tests per day).
