Controlled pore glass (CPG) is a well-established solid support for solid-phase oligonucleotide synthesis, but conventional CPGs are often designed with pore sizes and loadings that are not optimised for modern manufacturing. As oligonucleotide length and synthesis scales evolve, sub-optimal pore architecture and surface loading can limit productivity, reduce efficiency, and introduce variability.
PrimeMax™ CPG is a next-generation controlled pore glass engineered to deliver predictable, high-productivity performance for oligo synthesis. It combines optimised pore size, controlled surface area normalised loading (SANL), and next-generation surface chemistry to maximise usable synthesis scale while maintaining product quality.
Our first iteration, PrimeMax™ siRNA CPG is designed with reduced pore diameters (typically ~360-440 Å) relative to conventional CPGs, increasing accessible surface area and enabling higher effective loading without the negative effects of overloading. Loading is specified based on surface area rather than mass, ensuring consistent utilisation of reactive sites and avoiding efficiency losses associated with excessive surface density.
The solid support is manufactured using LGC Biosearch Technologies’ next-generation Native CPG process and incorporates trimethylsilyl (TMS) end-capped LCAA chemistry to enhance stability and minimise side reactions during synthesis. Comparative synthesis studies demonstrate that PrimeMax™ siRNA CPG delivers significant improvements in net full-length product yield and overall productivity versus incumbent CPGs, while maintaining comparable crude and purified impurity profiles.
