Lyo-Ready EpiScript™ Thermostable Reverse Transcriptase is a third-generation MMLV variant with reduced RNase H activity and enhanced thermostability compared to EpiScript RNAse H- Reverse Transcriptase. It supports first-strand cDNA synthesis at up to 60°C for higher specificity and can be used in one-step RT-qPCR with a compatible Hot-start Taq DNA Polymerase or qPCR master mix. This glycerol-free formulation is suitable for lyophilization.
Lyo-Ready EpiScript Thermostable Reverse Transcriptase is a General Purpose Reagent (GPR), labeled For Laboratory Use (FLU) in the United States as defined in CFR - Code of Federal Regulations Title 21. Outside the United States, the FLU statement is equivalent to Research Use Only. This product must be qualified and validated by clinical laboratory end users for suitability in the detection of any specific target using the procedures in this manual as guidance.
Figure 1. Amplification plots from Bio-Rad CFX96 real-time PCR for a dilution series with template RNA amplified using Lyo-Ready EpiScript Thermostable Reverse Transcriptase. Pre-lyophilization(green) and post-lyophilization(orange) overlaid.
Figure 2. Standard curves from Bio-Rad CFX96 real-time PCR for a dilution series with template RNA amplified using Lyo-Ready EpiScript Thermostable Reverse Transcriptase a) pre-lyophilization and b) post-lyophilization. Template levels are shown as Log Starting Quantity in copies per reaction. E is PCR efficiency with acceptable values between 90-110%. R2, slope and y-int are outputs of a linear regression analysis of the data.

Figure 3. Results from transcription times of 1, 5, 10 and 15 minutes using Lyo-Ready EpiScript Thermostable Reverse Transcriptase in a 4-plex RT-qPCR assay with a 60˚C reverse transcription temperature. This assay was run on a Bio-Rad CFX96 real-time PCR instrument. Template levels are shown as Starting Quantity (SQ) in copies per reaction and individual targets are represented by the fluorescence detection channel (Fluor).
