Quasar® 670 is an indocarbocyanine which fluoresces in the red region of the visible spectrum. This compound is a direct replacement for Cy5™. Quasar 670 amidite is used for the 5' labeling of fluorogenic probes used in 5' nuclease assays, Molecular Beacons™, and other detection assays. This amidite does not contain a DMT protecting group and can only be added to the 5' terminus of the oligo or internally on a free hydroxyl. BHQ®-2 will quench the Quasar 670 moiety.

Product usage:

  • Synthesis conditions:

    Due to the mild cleavage and deprotection conditions, we recommend using fast deprotecting amidites. Prior to dilution ensure that all product is at the bottom of the vial. Dilute to the recommended concentration and mix thoroughly in sealed vial to ensure that all contents are dissolved. For optimal coupling efficiency, we recommend treating the amidite solution with 3-4 Å activated molecular sieves (10% v/v).

  • Dilution:

    100 µmol/mL

  • Deprotection conditions:

    Oligonucleotides labeled with Quasar 670 may be deprotected using concentrated NH4OH, although some degradation can occur. Deprotect the Quasar 670 labeled oligo in concentrated NH4OH for 8 hours at 25 °C or for 1 hour at 60 °C. The use of methylamine should be avoided.

  • Quasar 670 is light sensitive material. Prolonged exposure to light may cause photo bleaching. This product is susceptible to oxidation when exposed to air or stored undessicated.
